½ tsp of soil was incubated at room temperature for 5 days in 9 ml yeast-peptone-dextrose (YPD) (1% yeast extract, 2% peptone, and 2% glucose) broth containing chloramphenicol (30 μg/ml) and ampicillin (100 μg/ml). 10 µl of homogenised cultures were inoculated into fresh media for a further 2 day incubation. 100 µl of both 1:100 and 1:10,000 diluted cultures were plated onto YPD agar (1% yeast extract, 2% peptone, 2% agar, 2% glucose) and incubated for 5 days. Potential yeast isolates were chosen for further investigation.
